5-Amino-1MQ Overview
Mechanism of Action
5-Amino-1-methylquinolinium (5-Amino-1MQ) is a selective inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that methylates nicotinamide and depletes NAD+ and SAM (S-adenosyl methionine) pools. By blocking NNMT, 5-Amino-1MQ increases intracellular NAD+ and SAM levels, enhancing cellular energy metabolism and reducing adipocyte (fat cell) differentiation. It has been shown to shrink fat cells and increase muscle stem cell differentiation.

5-Amino-1MQ Research Studies
5-Amino-1MQ, a potent, selective, and cell-permeable inhibitor of NNMT
Neelakantan H, Wang HY, Vance V, et al. — Biochem Pharmacol (2017)
Identified 5-Amino-1MQ as a potent NNMT inhibitor that increased NAD+ levels, shrank adipocytes by 50%, and reduced diet-induced obesity in mice without affecting food intake.

How does 5-Amino-1MQ differ from NAD+ supplementation?
NAD+ supplementation provides the coenzyme directly. 5-Amino-1MQ works upstream by blocking the enzyme (NNMT) that degrades NAD+ precursors. Together they can provide complementary NAD+ restoration through different mechanisms.
Can 5-Amino-1MQ reduce fat without exercise?
Animal studies showed 5-Amino-1MQ reduced body fat without changes in food intake, suggesting a direct metabolic effect. However, combining with exercise and diet optimization will produce the best results.
