FOXO4-DRI Overview
Mechanism of Action
FOXO4-DRI is a D-retro-inverso peptide that disrupts the interaction between FOXO4 and p53 in senescent cells. Normally, FOXO4 sequesters p53 away from mitochondria in senescent cells, preventing apoptosis and allowing these “zombie cells” to persist and secrete inflammatory factors (SASP). By blocking FOXO4-p53 binding, the peptide releases p53 to trigger selective apoptosis of senescent cells while leaving healthy cells unaffected.

FOXO4-DRI Dosing Protocol
Senolytic Protocol

What are senescent cells?
Senescent cells are damaged cells that have stopped dividing but resist dying. They accumulate with age and secrete inflammatory molecules (the SASP — senescence-associated secretory phenotype) that damage surrounding tissue, contributing to aging, chronic inflammation, and age-related disease.
Is FOXO4-DRI safe for healthy cells?
The FOXO4-p53 interaction that FOXO4-DRI disrupts is specifically upregulated in senescent cells. Healthy cells do not rely on this interaction for survival, so they are not affected. The original Cell paper demonstrated selectivity for senescent cells in vivo.
