GDF-8 Overview

Mechanism of Action

GDF-8 propeptide is the naturally occurring inhibitor of myostatin (Growth Differentiation Factor-8). Myostatin is a TGF-β family member that powerfully inhibits skeletal muscle growth. The propeptide binds and sequesters active myostatin, preventing receptor signaling. This releases the brake on muscle protein synthesis, satellite cell activation, and muscle fiber hypertrophy.

What happens when myostatin is blocked?

Belgian Blue cattle (myostatin mutation) exhibit extreme muscularity. A human child with the mutation showed extraordinary muscle development. Pharmacological inhibition aims to partially replicate this.

Is myostatin inhibition safe long-term?

Complete genetic knockout is well-tolerated in animals. Partial pharmacological inhibition shows acceptable safety in trials but long-term human data is limited.
